Tirzepatide is a 39-residue peptide with noncanonical residues and a C20 fatty diacid attached through a linker. That lipidated side chain contributes to albumin binding and prolonged exposure. An analysis that confirms only the peptide backbone is incomplete.
Orthogonal identity is essential
Intact LC-HRMS can support overall mass, but isomeric sequence errors, attachment-site mistakes and closely related conjugates may survive an intact-mass screen. Peptide mapping, diagnostic fragments and comparison with a qualified reference material strengthen assignment.

Sample handling can create the impurity
Lipidated peptides can show adsorption, solubility and carryover problems. Diluent, vial surface, needle wash, temperature and hold time should be qualified. A disappearing main peak may reflect poor recovery rather than chemical degradation; a late carryover peak may contaminate the next injection.
Why a vendor chromatogram is not enough
A chromatogram without method, wavelength, integration rules, system suitability and sample basis cannot establish identity or content. “Greater than 99%” should be treated as one result within a larger release package, not a complete quality conclusion.
